Efficacy Evaluation of Pinus sylvestris var. mongolica on Mice with Gastric Ulcers Induced by 150 mM HCl/60% Ethanol

Abstract
Pinus sylvestris var. mongolica (Pinaceae) is known to be rich in phenolic compounds which have antioxidant and anti-inflammatory properties. However, the underlying mechanism of action of the compounds has not been clearly elucidated. In this study, we investigated the effects of Pinus sylvestris var. mongolica (Pinaceae) water extract (PS) in a common ICR mice model with gastric ulcers (GU) induced by 150 mM HCl/60% ethanol. The mice were divided into 4 groups (n=8): Normal (normal group), Control (GU control group), SC (GU£«sucralfate 10 mg/kg), PS100 (GU£«PS 100 mg/kg), and PS200 (GU£«PS 200 mg/kg) groups. Ninety minutes after drug administration, the mice were orally administered 150 mM HCl/60% ethanol. The PS treatment significantly suppressed the level of malondialdehyde in the tissue and significantly downregulated the expression of NOX2 and p22phox. Moreover, it effectively upregulated the antioxidant enzymes, such as heme oxygenase-1 and superoxide dismutase through the activation of the Nrf2/KEAP1 signaling pathway. Besides, the PS-treatment significantly downregulated the expression of inflammation-related cytokines, including tumor necrosis factor-alpha and interleukin-1 beta, via NF-¥êB p65 inactivation through the AMPK/Sirt1 pathway. The results of our study thus demonstrated that PS has a significant effect in preventing gastric ulcers that occur due to decreased oxidative stress defense and inflammation.
